Hi all,
Aldehyde fixation involves reaction with primary aliphatic amines as
far as I know. However, I am less clear about any possible reactions
with aromatic amines. Our aim is to measure levels of an aromatic
amine in pfa-fixed cells by triple-quadrupole MS and we see a marked
reduction compared to unfixed cells. Could pfa be the reason?

Yes! I thought I had replied to you earlier. Anders Bjorklund and
others
have shown PFA and other aldehydes yield a fluorescent product with
catecholamines and indoleamines. This will obviously alter the MW you
are
looking for. If you search, one of the papers actually demonstrates the
reaction. You should be able to calculate the weights from that paper.

Thanks to Ed O'Connor's reply I delved into some really orginal work
that started in 1962. I found a very nice personal account of these
early experiments here:

Unfortunately, this method is not really what I was after as it is
about primary amines like dopamine and noradrenaline condensing with
formaldehyde (gas) to form fluorescent isoquinolines. Still, very
interesting reading.

Several people have used that approach not only to observe the
distribution in the brain but also to extract and estimate the
concentrations.
