Dear All,
I have a problem with my oral bolus sulfametoxazole chronopharmacokinetics studies in male rabbit (1,8-2,1 kg) the sampling time was done in 9 am. and 9 pm. the results was no significant differences (using t test statistic) between halflife and K elimination, the significant differences only happened on AUC 0-24 hour. sampling time points was 0, 15, 30, 60, 90, 120, 240, 360, 540, 720, 1440 minutes. My formula was combination between sulphametoxazole and trimetoprim, i am using bratton marshal assay. this is my result from my experiments
Pharmacokinetics parameter
9 am* 9 pm
Cmax (ug/ml) 15,956 +/- 14,34 14,369 +/- 6,60
T max (minutes) 34 +/- 2,55 58,20 +/- 0,84
AUC# (ug/ml.minutes) 2674.571 +/- 452,66 4536,532 +/- 1534,37
t 1/2 (minutes) 59,83 +/- 17,90 110,15 +/- 48,58
Kel /minutes 0,0123 +/- 0,0032 0,011 +/- 0,008
my literature in doing this experiments (oral administration of sulfametoxazole in rabit, using hplc) stated that pharmacokinetics parameter are the same result with human pharmacokinetics for example in halflife, the halflife value are 11 hour in am and 15 hour in pm.
I would appreciate it if someone could give me the reason of the differences between pharmacokinetics parameter in my result and in previous study.
Thanks,
