i want to extract drug form plasma. i tried number of times but I'm
unable to find the drug in plasma. could you suggest me how to exract
the drug. structure of my drug is
NH2CH2CH2NH CH2CH2S C6H5. 2HCl.H2O

One possibility is that your drug is metabolized in plasma (before
extraction) through oxidative deamination of the terminal methylene
amine
group to form an aldehyde and ultimately a carboxylic acid metabolite.
The reaction can be fairly fast, especially in human plasma. The enzyme
responsible for such a process is SSAO (semicarbazide sensitive amine
oxidase). If this is indeed what's going on, you can stabilize the
compound
in plasma with hydralazine.

First add different concentration of your test compounds into blank
plasma, to see at which concentration level, you could get recovery.
By the way, what kind of extraction method have you used. Maybe you
need adjust the pH value to get better recovery.

Shows a primary and a secondary amine. Is this the structure? Have
you checked the stability and recovery from aqueous solution? what pH
and what method of extraction have you tried (cation exchange?)

Dear Pankaj
Can you tell me what is LLOQ and also how much plasma volume your
taking? Which extraction method your following. Hope it is liquid-
liquid extraction. You just add 0.1% ammonia try to extract with
ethyl acetate or TBME.
