Dear All,
I look for examples of drugs (chemicals only) with terminal half-lives > 48 hours.
Kind Regards,
Roberta

Roberta,
I believe rapamycin (sirolimus) has an elimination half-life of about 60 hours.

Hi Roberta
I believe amiodarone has the distinction of having the longest
terminal elimination half-life amongst marketed drugs (mean t1/2 = 58
days after a single-dose). Efavirenz has a terminal half-life of
52-76 hours after a single dose and 40-55 hours after multiple doses.
Hope this helps.

moxidectin and amiodarone also have long half-lives.

Roberta,
Amiodarone has a half-life of about 50 days, which is exceptionally very prolonged.

The mean elimination half life of Aripiprazole
1. In extensive metabolizers is 75 hours
2. In poor metabolizers is 146 hours
Approximately 8% of Caucasians lack the capacity to metabolize CYP2D6
substrates and are classified as poor metabolizers (PM), whereas the
rest are extensive metabolizers (EM). PMs have about an 80% increase in
aripiprazole exposure and about a 30% decrease in exposure to the active
metabolite dehydroaripiprazole compared to EMs, resulting in about a 60%
higher exposure to the total active moieties from a given dose of
aripiprazole compared to EMs.
