Hi there,
I have a question regarding magnesium chloride concentration in CYP-mediated in vitro metabolism assays. Does it matter if we use 3.3 mM (which is the concentration in the BD Biosciences NADPH regenerating system) or higher (e.g. 13.3 mM). Because recently one of our graduate students got higher metabolite formation (at least 2 fold) with higher concentrations of magnesium chloride (3.3 mM Vs 13.3 mM) in the system.
Your input will be much appreciated.

It's a good observation, but I have a question that it's happened with all
type of CYPs or only few. Please mention that. We are doing lot of validation
assays with Recombinant CYPs which expressed from yeast in our lab. So
definitely I will do MgCl2 variation assays and I will come back with the
results.
