Dear friends,
I plan to use Caco-2 monolayers in experimental predictions of intestinal
drug transport. Is there any passage-dependent effect for this cell line
such as diminished functional expression of intestinal transporters,
growth rates and ability to differentiate at high passages? If there is at
what passage range?
Thank you.
Best regard,
swtan

Hi Swtan
You can find lot of articles in Journal of Pharmaceutical sciences regarding the effect of passage numbers on Caco-2 permeability. I believe people had used the cells till passage number of 80-90, but i would suggest you to go maximum about 30-45 passage number only to ensure cells integrity and functional aspects.
Experts comments are welcome.....

Khalid
