Hey Fellow Scientists
Need some technical inputs. Anyone on this forum ever have done sampling of CSF (Terminal collection) from mouse by direct ciserna-magna puncture or by any other method? If yes, I would really appreciate you sharing the technical aspects of the procedure.

We are regularly sampling CSF from anesthetized rats in our lab, its quite easy and fast. But mouse I believe would be bit difficult to perform, further volume you may get is at max 10-20 uL. FYR go through the following video protocol published in JOVE. Author has described an invasive technique for CSF sampling from mouse.

This is counter to all of my training, which would say that at 3 significant
digits, 10600, or 1.06 E4, would mean a value between 10550 and 10649 (or
1.055 E4 to 1.064 E4). There is no way it would imply an accuracy to the 6th
significant digit!

You can try this method for CSF collection:
First anesthetized the rat (you can use Isoflurane). A cannula then assembled by inserting a 26G hubless needle into one end of a 12" length of PE-10 tubing. Rats will be then positioned in sternal recumbency. A small incision will be then made in the skin above the atlanto-occipital area, and the head will flexed downward at approximately 45o.
The CSF will be collected by direct needle puncture into the cisterna magna, using the occipital bone and the wings of the atlas as landmarks.
Color change in the sample will be monitored just above the needle during collection. Upon observation of color change, the PE tubing will be quickly clamped off above the color change and cut just above the clamped site. The non-contaminated sample will draw into the syringe.
