Dear all,
I will run a study in rats, which is requested 1.6 mL/kg dose volume injected in one site. It is out of the recommended range for IM dose (0.1-0.2 mL per site). I would like to know if anybody have the experience with such large dose volume in rats? Or may anyone suggest a scientific publication on overview of the effects of the large dose volume on animals.
Any information would be much appreciated.
Many thanks,
Yuming

Dear Sir,
In book titled " Anesthetic toxicity" by Susan A.Rice, Kevin J.Fish, the dosage volume guidelines recommendation in rat is given as follows (Page 29)
Ideal volume : 0.2 ml/kg
Limit volume : 2.0 ml/kg

In a research article titled " Dose-Dependent Amelioration of Gentamicin-Induced Nephrotoxicity in Adult Swiss Albino Rats by Vitamin B-complex -A Preliminary Study " published in Tropical Journal of Pharmaceutical Research, Vitamin B complex was given intramusculary at the dose of 1.5 ml/kg to Adult, female, Swiss albino rats, weighing170 +/- 20g.

Overview of effects on large volume dose administration in animals:
Administration of excessive dose volumes may produce pain, excitement, altered physiological parameters (e.g., serum electrolyte imbalance, increased blood pressure, increased respiratory rate, etc.), and cause abnormal compound absorption...
