Dear Group,
I am rather new to this program and modeling in general. I am trying to simulate a study in TS using a 1 compartment model.
I have fit my individual data in winnonlin and calculated the individual values for the parameters: ka, V, ke.
At this point I am not up for tackling NONMEM and just want to simulate a study using those values.
In my computation box, I have Vc mean, Vc eta, and Vc expression - Vcmean *exp(Vceta) And the same for the other parameters.
What I am confused about is how to enter the values in the block for Vcmean and Vceta.
I am assuming that my data is lognormal for Vc. So first, should I log transform data first then enter the mean in the box and list the type as lognormal. Or do I enter the mean as a raw value then list type as lognormal? Or are neither of these correct? Likewise I have a similar question for how to handle the standard deviation box? Note, I want to avoid having negative values. Also, I put the variable at study level.
Next question is for the eta box. I put this at subject parameter for level. Again do I transform the data or use untransformed data to enter mean and sd for eta. Again I am confused about this one, but I am assuming eta has a mean of zero with a sd. However, if you enter lognormal distribution you get an error because zero is not allowed?
Again I realize there are many complexties etc. but at this stage I am only trying to do a basic simulation to generate some simulated data similar to actual data. My goal is to get something working before doing something more advanced.
