Dear all,
I would to have a model to study the drugs acting on sodium current without the electrophysiological methods.
I have tried to use the commercial sodium binding fluorescent indicator (SBFI) in SH-SY5Y cell line.
I have read the fluorescence signal at the spectrofluorometer equipped with fast filter for a ratio dye lecture. I have a small but stable signal; I am able to calibrate a signal.
When I try to use veratridine (1-100 micromolar) I look a decrease of fluorescence signal. Maybe the reason is the fluorescence signal of veratridine molecule "per se". If I use the veratridine concentration that don't give a decrease of a signal I don't look the difference versus the control. Maybe I have too small change of sodium current.
Do any of you give me some advices?
