Hi
I have a very basic question regarding permeability assay.
Which cell lines generally people used means:
MDCK 1 OR MDCK II OR MDCK wild type.

If anyone want to do permeability assay using cell lines, Caco2 cell line is the first choice. It is most commonly used all over the world to study permeability of NCEs. Most common issue associated with Caco2 cell line is time taken to propagate the cell line. It takes 21 days to propagate. To over come this issue, many people prefers MDCK cell line to study the permeability which takes 4-5 days to attain full confluency. Wild type MDCK do not have MDR1 gene which is responsible for efflux of the drug. If your aim is to study bi-directional permeability then MDR1-MDCK is more appropriate. Nowadays, people have come up with shorter duration Caco2 cell line also (Rapidly growing Caco2 cell lines).
