Dear PharmPK users,
I would like to perform a PK/PD with data from a model of collagen-induced arthritis in mice.
In this model, animals were sensitized and three weeks later were boosted with another collagen injection to induce disease. Seven days following this injection the animals were treated with the test compound at different dose levels. The drug was administered orally bid for 21 days. During this period the mice were scored every 2-3 days for signs of arthritis in each paw. Upon completion of the study blood samples were taken for analysis of the drug levels.
Arthritic scores were as follows:
Day 0: around 4.5 for all groups
Day 21:
Control 12.0
Low dose 10.25
Mid dose 8.57
High dose 2.43
PK of the drug was adequately described by a 2-compartment model. My problem is with the PD part. What type of model would be suitable for this type of data? Maybe an indirect effect model of inhibition of input?
Any suggestion will be greatly appreciated.

Hi Angela,
There are several publications on the PK/PD modeling of the animal model you describe by Dr. Jusko's group. A search on Pubmed using Jusko wj + collagen will give you 5 articles to start with.
