Hello.
I am having problems validating an S9 Phase II stability assay. Unfortunately, 7-Hydroxycoumarin can't be detected by our LC-MS/MS device. Does anyone know another reference drug with a reasonably high S9 clearance, that is mostly metabolized by conjugation (glutathione-, acetyl-, sulfo- or glucuronide-), and not oxidated by CYPs? I am also looking for reference clearance data.

My response to your query is on detection of 7-hydroxycoumarin. We had
analyzed this compound in on of our lab successfully. The polarity we
used was negative ionization with turbo ionspray source. The MRM
transitions we monitored were 7-hydroxycoumarin 229/153 and if you are
looking to monitor glucuronide conjugate, 7-hydroxycoumarin-Gluc
was 405/153. The mobile phase system was 0.2% formic acid in water
(aqueous) and 0.2% formic acid in ACN (organic).
