Hi everyone,
Thanks for the thread. I am also trying to extract my drug from feces and I tried some recovery
experiments using feces as a matrix. I tried different protocols till date and could not see the
peak of the drug even in recovery experiment. At last after heating the dry feces I got at most 20%
recovered feces. Could you please let me know any other method I can try to enhance the recovery of
the drug??
My compound is a glycosylated compound which is water soluble. For extraction solvent I tried
different solvents like butanol, Ethyl acetate, Methanol etc., but only in ACN worked better
compared all the solvents.

Hello Sumit Basu,
I would like to know the nature of your analyte (Polar, No-polar, pKa etc). This will help to select
suitable solvent and extraction techniques.
In general:
You need to first homogenate your feces using water or any other suitable buffer (1:3 ratio). Then
you can try protein precipitation (PPT) using ACN like say (25 µL of homogenated fecal sample and
add 100 µL of ACN) vortex for ~5-10 min. Centrifuge for 4000 rpm for 10 min. Take ~60 µL supernatant
for LC-MS/MS analysis.
You can also try SPE, best work for urine and fecal sample analysis.
