Dear colleagues/students,
Would a total body clearance (CL total) of 1.23 L/Kg/h after iv-bolus
dose of a drug to an on average 280 g, male SD rat, be considered a
high, low or medium clearance?
Can anyone give me a suggestion?

Concerning the question if CL total of 1.23 L/Kg/h after iv-bolus

It is important to remember that ClB is related to extraction ratio and
blood flow (cardiac output). Then, taking account of the cardiac output for
a rat of 0.2 g and considering critical values for extraction ratios
high>0.7 and low <0.3 :
ClB is high if higher than 85 ml/kg/min (5.1 l/kg/h)
ClB is low if lower than 37 ml/kg/min (2.22 l/kg/h)
I would say that for your rat a ClB of 1.23 l/kg/h should be considered low.

Hi Panteha,
Are you referring to plasma or blood clearance? If it=B4s blood, then
you are dealing with hepatic clearance and for a 250g rat that is
approx. 4 L/kg/h
