I've been assaying tobramycin in sputum (made up of DNA and mucin) using
an HPLC techniques which involves derivatization with phthaldialdehyde
followed by fluorescence detection.
I first hydrolyse the sputum in sodium hydroxide which has the effect of
solubilising all the "tissue" components. I then derivatize and assay
directly.
