[I seem to remember some researchers suggesting a continuous
reverse-infusion (i.e. continuous drawing of blood) to a pooled
sample that was then assayed (one assay) to get the AUC. Probably
about 10 years ago now...forget the name and not sure what they
called it. Maybe someone on the list can help with more details - db]
Dear group,
I would like to know the pros and cons about the practice of PK screening
by pooling 6-8 equal volume blood samples over a period of time and
assaying the drug just to get the AUC without actually generating "the
curve". This way I suppose you can save time and cost by rejecting the
unsuitable ones early on and for the promising ones you can go back and do
the studies traditional way. If anybody is doing this type of screening I
like to know what you think about this.
Thanks for your help.

Dear Chandrani Gunaratna
I remember reading a paper on assaying pooled plasma to
estimate Css,ave (hence AUC). I think the following
reference describes the process.
Hop C et al. Plasma-pooling methods to increase throughput
for in vivo pharmacokinetic screening. J Pharm Sci
1998;87:901-903
Hope this is helpful.
Regards
Steve
